Differences in OD600 Readings–Where Do They Come From

In microbiology, Optical Density (OD) measurements at 600 nm (OD600) are essential for evaluating cell culture density. However, OD600 readings for turbid samples, like cell cultures, primarily reflect light scattering rather than molecular absorption under the Beer-Lambert Law. Variations in spectrophotometer optical setups—such as distances and optics—can lead to differing OD600 values for the same sample.

To ensure comparability across instruments, normalization of OD600 readings is crucial. This can be achieved by correlating readings from different instruments using the same sample or by constructing calibration curves based on known cell concentrations. Instruments like the NanoPhotometer® or Implen OD600® include default correction factors for consistent results across setups.

For accurate measurements, we recommend using disposable cuvettes instead of micro/nano volume technologies. Cuvettes provide more reliable readings by minimizing variations in sample volume, which is critical for precise OD600 determinations in cell cultures. Adding glycerol to samples can stabilize suspensions, ensuring consistent OD readings over time.
